Skrining Anti Depresant Waltheria indica L. Dengan Menghambat Monoamine Oxidase Dan Uji Aktifitas Ektrak Etil Asetat

Depresi merupakan salah satu gangguan kesehatan mental yang prevalensinya semakin meningkat pada generasi muda dan memerlukan alternatif terapi berbasis bahan alam yang lebih aman. Penelitian ini bertujuan mengevaluasi aktivitas antidepresan ekstrak etil asetat Waltheria indica L. melalui pendekatan in vivo dan docking molekuler. Metode yang digunakan meliputi uji in vivo pada tikus Wistar dengan model Chronic Mild Stress (CMS) yang dievaluasi menggunakan Forced Swimming Test (FST) dan Open Field Test (OFT), serta analisis docking molekuler menggunakan AutoDock Vina terhadap target Monoamine Oxidase. Hasil docking menunjukkan senyawa quercetin memiliki energi ikatan paling rendah (-6,977) dengan interaksi residu kunci, menandakan potensi kuat sebagai penghambat. Uji in vivo memperlihatkan adanya penurunan imobilitas dan peningkatan lokomotor yang signifikan seiring kenaikan dosis, dengan efek terbaik pada dosis 350 mg/kgBB. Data ini konsisten dengan kontrol positif (Ashwagandha®) dan mendukung adanya mekanisme antidepresan. Kesemipulan ekstrak etil asetat W. indica berpotensi dikembangkan sebagai kandidat terapi antidepresan berbasis bahan alam.
